The marathon course consists of a 10km loop through the Parliamentary Triangle, followed by a doubled stretch along the southern and then northern shores of Lake Burley Griffin, from Bowen Park to near the Glenloch interchange. Participants will then cross Kings Avenue Bridge on the outwards journey, and Commonwealth Avenue Bridge on the return journey.

The Canberra Times Canberra Marathon gives participants the opportunity to go 'beyond the marathon' and compete in a 50km Ultra Marathon. After finishing the 42.195km marathon and being recorded as a marathon finisher, participants are eligible to continue and complete the Canberra 50km Ultra Marathon. There is no need to pre-register for this bonus event, all 2012 full marathon participants are eligible to compete in the Ultra Marathon on event day.
